There will be more than 240 stallholders taking up to 400 car park spaces at Hunter Stadium.
Cars, tents and tables will be full of treasures and treats from the quirky to the creative, including handmade goodies, vintage clothing, antiques, restored furniture, baby items, books, sunglasses, collectables and car parts.
The sale is also a major fund-raiser for Hunter Life Education. The target for this year is to raise $25,000.
Life Education aims to help young people stop illegal drug use, reduce smoking, avoid or delay alcohol use and make healthy choices about their diet and lifestyle.
In the past year, 32,0000 Hunter children have attended its programs.
Hunter Life Education executive officer Sally Coddington said the organisation valued the support of stallholders and the general public.
She said the event was a great way to get people involved in something that not only benefited Hunter Life Education, but the wider community.
"In its ninth year, the World's Biggest Car Boot Sale has become an important feature on Newcastle's annual event calendar, and this event will not disappoint," she said. "Everyone is sure to pick up something special at a great price."
There will also be food and drink, free entertainment and appearances by Healthy Harold and NBN's Big Dog.
Entry is a gold coin donation and all proceeds go to Hunter Life Education.
