The Port to Port MTB is the sister event to the successful Cape to Cape MTB, which has been well received by West Australians since it was first held there six years ago.
The four-stage event will be held in Newcastle and the Hunter Valley from Thursday, May 29, to Sunday, June 1.
It will start at Nelson Bay Marina, where riders will be greeted by sandy tracks, fire trails, testing hill climbs and the steep descents of the Hunter Valley.
The race will end with "Super Sunday", testing riders on the world-class Glenrock MTB trail.
With a strong selection of elite riders, including Cape to Cape MTB champions Jenny Fay and Andy Blair, event organisers said it was sure to be a tight scramble to the end.
