Event director Jason Becker said there would be a back-to-school dress-up theme, and Wangi Men's Shed would cook up a post-run barbecue with all funds raised going to Cure Cancer Australia.
The run starts at 8am and organisers are hoping to break the 600 mark for participants.
Numbers have grown since it began in 2013 with Mr Becker reporting attendance hovering between 380 and 420 on a weekly basis now.
Parkrun is a free, five kilometre run where participants run, walk or jog for their own enjoyment.
The Lake Macquarie event takes place at 1a First St, Booragul, opposite Booragul Primary School.
Other Parkrun events are held throughout the Hunter with the Newcastle Parkrun taking place in the area surrounding Throsby Creek every Saturday morning from 8am.

Meanwhile, the Maximum Adventure Race Series will return to the shores of Lake Macquarie on Saturday, February 16, launching from Cams Wharf.
The race combines mountain biking, trail running and kayaking.
The event is designed for teams of two people and has a classic course and a novice course to choose from.
